The roots of Millettia speciosa is used as a medicinal herb to support fortification and increase the body's resistance. This study has a purpose optimal extraction condition of total saponin triterpenoit and polysaccharide from roots of Millettia speciosa with. Using response surface methdology (RSM) and experimental according to Box - Benhken design has optimized extraction process total saponin triterpenoit content (TSTC) and polysaccharide content (PC) with three factors: ultrasonic power (X1), time extraction (X2) and ratio of water raw material (X3). Folow this model, the optimal parameter for extraction process to obtain the highest content of total saponin triterpenoit and polysaccharide is ultrasonic power of 375 W, time of 65 mins and ratio of water raw material of 38 mL/g. The experimental values of TSTC and PC were 1.25 ± 0.02 mg/g and 7.86 ± 0.03 mg/g.
